Retatrutide Phase 3 Update: September 2026 Publications Explained
What the September 29 TRIUMPH-1 and TRIUMPH-2 releases add to the evidence, how to interpret the headline numbers, and where to find the original sources.

What is new in the September 2026 retatrutide news?
A topline announcement presents selected findings before the full paper. A peer-reviewed publication gives readers a more detailed basis for evaluating methods, endpoints and interpretation. The September releases should therefore be understood as a publication milestone, rather than the first evidence that these trials met their objectives.
- TRIUMPH-1: the obesity paper was published online on September 29. [1]
- TRIUMPH-2: detailed findings in participants with type 2 diabetes were released on September 29. [2]
- Earlier reporting: Lilly announced TRIUMPH-1 topline findings on May 21 and TRIUMPH-2/TRIUMPH-3 topline findings on July 23, 2026. [3] [4]

TRIUMPH-1 and TRIUMPH-2: key results in context
TRIUMPH-1 randomized 2,339 adults without diabetes and evaluated outcomes over 80 weeks. The published weight endpoint used a treatment-regimen, intention-to-treat analysis. [1]
| Trial and analysis | Reported finding | Comparator |
|---|---|---|
| TRIUMPH-1: intention-to-treat | 25.0% mean body-weight reduction in the 12 mg study arm | 3.9% with placebo |
| TRIUMPH-1: earlier efficacy estimand | 28.3% mean body-weight reduction in the 12 mg study arm | 2.2% with placebo |
| TRIUMPH-2: efficacy estimand | 20.8% mean body-weight reduction in the 12 mg study arm | 4.0% with placebo |
Sources: TRIUMPH-1 publication and Lilly releases. Study-arm labels describe trial groups, not instructions for use. The two trials enrolled different populations, and the rows use different estimands. [1] [2] [3]
What else did the publications examine?
TRIUMPH-1 also assessed knee osteoarthritis pain and obstructive sleep apnea in defined subgroups. Those findings must be interpreted within their specific populations and analysis plans. [1]
TRIUMPH-2 enrolled 1,152 participants with type 2 diabetes and obesity or overweight. Lilly reported an average A1C reduction of 1.5 percentage points in the 12 mg group, compared with 0.2 percentage points with placebo, using the efficacy estimand. [2] Our retatrutide type 2 diabetes research guide covers the broader topic.
Why do retatrutide headlines show both 25.0% and 28.3%?
The figures answer different statistical questions. An estimand defines the treatment effect a trial seeks to estimate, including how events such as treatment discontinuation or additional treatment are handled.
- Treatment-regimen estimand: evaluates the assigned treatment strategy regardless of adherence or prohibited weight-management treatment.
- Efficacy estimand: estimates outcomes under the assumption that participants remained on the study intervention without prohibited weight-management treatment.
Lilly reported both analyses in its May release. The 28.3% headline was not a separate trial, and the September publication's 25.0% result does not mean the trial was repeated with a weaker outcome. [3]
This also limits comparisons with other compounds. The September results do not establish superiority over another medicine in a direct comparison. Read our retatrutide vs tirzepatide, retatrutide vs semaglutide and retatrutide vs CagriSema guides with those differences in mind.
Safety findings, limitations and approval context
Gastrointestinal adverse events were the most common in TRIUMPH-1. [1] In TRIUMPH-2, Lilly reported discontinuation due to adverse events in 7.7% of the 12 mg group and 4.9% of the placebo group. [2] Efficacy findings should always be read alongside adverse events, discontinuations and missing-data methods.
These studies do not establish that every population will respond similarly, that rare risks have been excluded, or that results transfer to independently sourced research materials. A clinical trial evaluates a defined investigational product under controlled conditions.
Lilly's September announcement describes retatrutide as investigational and states a planned U.S. application in the first quarter of 2027. An intended submission is not regulatory approval. What Canadian researchers should verify beyond the news
Clinical evidence and supplier documentation answer different questions. A journal article describes a study. A certificate of analysis describes the tests performed on a submitted sample. Neither substitutes for the other.
For laboratories evaluating where to buy peptides in Canada, useful checks include the compound identity, reported purity, analytical methods, lot identifiers and the relationship between a report and the material supplied. A high chromatographic purity figure alone does not establish sterility, absence of endotoxins, clinical equivalence or suitability for human use.
Luxara Labs provides public lab results and COAs. Use the COA interpretation guide, peptide purity standards, Transparency Hub and Luxara Labs Quality Standard to examine what the documentation supports.
Further context is available in the Peptides Canada hub and Canadian research-use regulations guide. The Retatrutide laboratory research product page is a separate commercial resource, not evidence that Luxara materials were used in Lilly's trials.

Research quality, traceability and supplier accountability
Useful documentation should help a laboratory move from a product listing to the relevant analytical report, understand what was tested and identify who can answer questions. These resources support that review without substituting for the primary clinical literature.

Retatrutide Phase 3 update FAQ
What was the exact release date?
The TRIUMPH-1 paper and Lilly's detailed TRIUMPH-2 announcement were released September 29, 2026. [1] [2]
Was September 29 the first announcement of these trial results?
No. TRIUMPH-1 topline results were announced May 21, 2026. TRIUMPH-2 topline results followed July 23. [3] [4]
Why should I check the estimand?
It tells you which treatment effect a reported number estimates. Results based on different assumptions should not be treated as interchangeable.
Do these results validate a supplier's research vial?
No. Trial findings do not authenticate an independently supplied vial. Material verification requires its own analytical documentation and traceability.
Where can I find the original research?
The primary sources are listed below, including the NEJM paper, Lilly's dated announcements and trial registry records.
